Adult Congenital Heart Association Celebrates 20th Anniversary

Philadelphia, Pa., Feb. 22, 2018 (GLOBE NEWSWIRE) -- The Adult Congenital Heart Association (ACHA) is proud to announce its 20th anniversary during Heart Month, in recognition of two decades of advocating for lifelong care for those with congenital heart disease (CHD). Throughout the year, ACHA will celebrate the significant strides that have been achieved to enable individuals born with this most common birth defect—diagnosed in one in 100 births—to live happy, healthy adult lives.

ACHA is the only non-profit in the country dedicated solely to the unique social, educational and medical needs of the 1.4 million adults in the U.S. with one of the many different types of congenital heart defects. Karen Klein McNulty and two friends who were also living with CHD—Anthony Cordaro, Jr. and Bonnie Paulsen—founded ACHA in her parents’ living room as they sought a shared network for support and information. The group’s goal was to form a community where they were not alone, living with a disease that was largely invisible to the world. Today, ACHA offers its growing membership a wide range of resources focused on education, outreach, advocacy, and promotion of research related to CHD.

“This is a significant milestone for ACHA; thankfully, due to medical advances, more children with CHD are growing into adulthood,” said Mark Roeder, President and CEO of ACHA. “Through our advocacy and awareness campaigns and, most recently, the accreditation program we launched, we are prioritizing the need for constant support and specialized care to meet the needs of adults with CHD. We look forward to continuing to extend our impact in the years to come.”

Throughout these 20 years, ACHA boasts a number of accomplishments, most notably, serving the needs of more than one million adult patients with CHD. The organization has grown to include a Medical Advisory Board made up of top adult congenital cardiologists throughout the U.S.; the Heart to Heart Ambassador Program, which connects ACHA patient and family members with trained peers who provide support, guidance and resources; and the ACHA Actelion Fellowship Award, which provides individuals pursuing certification and a career in ACHD care with multidisciplinary clinical and research training.

Advocacy and public policy are key functions of the organization. In 2010, ACHA helped draft the Congenital Heart Futures Act, which provides government funding for public health research and surveillance on the issues that children and adults with CHD face. The effort continues today as ACHA actively advocates for the Congenital Heart Futures Reauthorization Act.

Throughout 2018, the organization plans to engage the broader community by co-hosting 18,000 participants in Congenital Heart Walks in 25 U.S. cities to benefit both ACHA and The Children's Heart Foundation. ACHA is also planning regional conferences in Northern California, Hollywood, FL, and Spokane, WA, along with inaugural gala celebrations in Los Angeles, Denver, Houston, and Boston.

Earlier this year, ACHA launched the ACHA Adult Congenital Heart Disease (ACHD) Accreditation Program, a targeted initiative to build a community of support and network of experts with knowledge of the disease. ACHA worked with doctors, physician assistants, nurse practitioners, nurses, and ACHD patients to define and vet the accreditation criteria and process. To date, there are 17 accredited centers in cities throughout the United States: Los Angeles, CA; Palo Alto, CA; Aurora, CO; Hollywood, FL; Boston, MA; Ann Arbor, MI; St. Louis, MO; Omaha, NE; New York, NY; Cincinnati, OH; Columbus, OH; Charleston, SC; Dallas, TX; Houston, TX; Seattle, WA; Spokane, WA; and Milwaukee, WI.

From Genes and Cells to Saving Lives: NEJM Reports Positive Results for Larotrectinib against TRK-fusion Cancers

Basic science from University of Colorado Cancer Center leads to clinical trial results reported in New England Journal of Medicine: Of 55 patients with TRK-fusion cancer who progressed after standard-of-care treatment, 44 responded to larotrectinib, with 38 remaining on the trial or receiving surgery with curative intent at median 9.4 month follow-up.

AURORA, Colo. (PRWEB) February 22, 2018

In 2013, the labs of University of Colorado Cancer Center investigator Robert C. Doebele, MD, PhD, and Dana-Farber Cancer Institute investigator Pasi A. Jänne, MD, PhD reported in Nature Medicine the presence of TRK gene fusions as oncogenic drivers in patient samples of non-small cell lung cancer. Now five years later, the New England Journal of Medicine has published results of three early studies of the drug larotrectinib (formerly LOXO-101) to treat advanced-stage cancer patients whose tumors harbor these TRK fusion genes.

“The story of this drug demonstrates a new paradigm in anti-cancer drug development. We have discovery and validation of a new oncogenic driver, creation of an assay to find which cancers harbor TRK fusions, drug design and preclinical testing, and now academia-industry partnership that produces very promising clinical results,” says Doebele, director of the CU Cancer Center Thoracic Oncology Research Initiative and co-author of the current study.

The three clinical trials looked for TRK fusion genes in patients whose cancers had progressed after standard-of-care treatment (a phase 1 study involving adults, a phase 1–2 study involving children, and a phase 2 study involving adolescents and adults). Overall, 55 patients ranging in age from 4 months to 76 years, representing 17 cancer types, tested positive for TRK fusion and were treated with larotrectinib. Overall response rate was 75 percent (44 of 55 patients). At a median follow-up of 9.4 months, 86 percent of the patients with a response (38 of 44 patients) were continuing treatment or had undergone surgery that was intended to be curative. No patients discontinued treatment due to adverse side-effects.

“This study also demonstrates the shift from defining cancers by the site at which they occur toward defining them by the molecular changes that drive their growth,” Doebele says. “In terms of treatment, these trials show that rather than seeing these as ‘lung’ or ‘colorectal’ or other site-specific cancers, we can think of these as ‘TRK-fusion cancers’ and treat them according to this driver.”

Larotrectinib is one in a class of drugs known as tyrosine kinase inhibitors (TKIs), that block the action of target genes. Previously approved TKIs including crizotinib and alectinib inhibit the action of oncogenic genes ALK and ROS1. Larotrectinib uses a similar strategy to inhibit the gene TRK, which is active during early embryonic development, should remain silent in adult cells, but can be improperly reactivated by fusion with partner genes. Doebele and colleagues originally received the drug for study from Array BioPharma of Boulder, CO, and have developed it in partnership with Loxo Oncology of Stamford, CT.

“I can’t tell you how gratifying it is to see our early lab work with genes and cells leading to a treatment that is literally saving patients’ lives,” Doebele says. “This is the dream of all scientists who choose to go into cancer research.”

Due to the durability of responses to larotrectinib, the current studies will continue to follow patients to determine how long the drug is expected to control TRK-fusion cancer. Future studies are planned. Reported by PRWeb 3 hours ago.

Klondex Increases Credit Facilities to $45 Million, Complimenting 2018 Cash Flow Initiatives; Company Management to Participate in the BMO Global Metals and Mining Conference

$

VANCOUVER, British Columbia, Feb. 22, 2018 (GLOBE NEWSWIRE) -- *Klondex Mines Ltd. (TSX:KDX) (NYSE American:KLDX)* (“Klondex” or the “Company”) today announced the Company has increased the borrowing capacity of its credit facilities. The Company also announced its participation in the BMO Capital Markets 27^th Global Metals and Mining Conference.  The Company has increased its revolving credit facilities by $10 million to $45 million. The increase includes an addition of $5 million in the revolving credit facility (the “Revolver”) to $40 million and an additional working capital tranche of $5 million. During the fourth quarter of 2017, Klondex utilized the Revolver to purchase 8,000 gold ounces at $1,269 per ounce and delivered them against its outstanding gold loan to Franco-Nevada, one year early.  The gold loan is now paid in full. Klondex has delivered a total of 38,250 gold ounces to Franco-Nevada since February 2014. Repaying the gold loan improved working capital by replacing a current debt with a non-current debt and will improve 2018 quarterly cash flow as it frees up 2,000 gold ounces per quarter. Additionally, the Company’s gold supply agreement with Waterton will expire on February 28, 2018. This agreement provides the option for Waterton to purchase gold produced from the Fire Creek Mine. Historically, the realized purchase price under this agreement has been at a discount to market price, which negatively impacted Klondex’s cash flow.

Barry Dahl, the Company’s Chief Financial Officer, said “It’s been a pleasure working with Investec Bank plc since 2016. Over the years, Investec has been flexible to structure the credit facilities to match the growth of the Company.”

Paul Huet, President and Chief Executive Officer commented, “These three financial items will all have a significant positive impact on our working capital flexibility for 2018.  Historically, Klondex’s focus has concentrated on growth and consolidating the mining district where we operate in Nevada. Our focus for 2018 is different. In 2018, we have refocused our efforts on generating cash. This emphasis was a contributing factor in the decision to place True North under care and maintenance, which should provide an opportunity for the Company to add cash to the balance sheet. As previously stated on December 21, 2017, Klondex will continue to utilize our core competencies and focus on narrow vein, high grade underground mining in Nevada. The Mars pit is a new discovery that we believe provides significant growth potential and future optionality for the Company.”

BMO Conference

Paul Huet, the Company’s President and Chief Executive Officer will participate in the BMO Capital Markets 27^th Global Metals and Mining Conference in Hollywood, Florida on February 28, 2018. The presentation will begin at approximately 8:45am Eastern Standard Time. A live audio webcast of the presentation may be accessed through the Investor Relations section of the Company’s website: www.klondexmines.com.

Scott Lipps's Modeling/Celebrity Agency "Lipps LA" (https://www.lipps.la/) takes home the coveted "Best of Los Angeles Award" for “Best Visionary Modeling Agency” 2018

LOS ANGELES (PRWEB) February 22, 2018

Scott Lipps, renowned modeling/celebrity agent and head of Lipps LA located at the Dream Hotel in Hollywood, has won the "Best of Los Angeles Award" for “Best Visionary Modeling Agency” 2018, adding it to his already long list of accolades. Describing his prominent new business, Lipps says, "we have constructed a vertically integrated agency, packaging content creation, model / celebrity management & audience building to create a one stop shop for talents & brands in the form of a digital agency."

When asked about his goals for the company, Mr. Lipps had this to say: "Honestly, for me it’s always about building the biggest entertainment-related company you can build. That’s always my ultimate goal in anything that I do. The models are first and foremost, and what I’ve done for 25 years. There’s a huge celebrity presence in LA, which is very exciting because I’ve represented a lot of celebrities over the years in New York, and this is where they are based. From a digital perspective, I would love to disrupt the business and do things differently. We’ll have the traditional model calling and the celebrity representation in the digital sense, but we’re also building out a whole side of things that aren’t done yet, including brand consulting and content development and working to market brands along with our talent, and packaging it together."

The “Best of Los Angeles Award” community was formed three years ago and consists of over 3,800 professional members living and working in Southern California. It celebrates the best people, places and things in Los Angeles with a slogan “No Ads. No B.S. Only the Best.” (https://www.facebook.com/BestofLosAngelesAwards/)

“The mission of the community is to celebrate the best of Los Angeles, and allow its community members to connect with other members who share the highest standards of quality and integrity,” said community spokeswoman Aurora DeRose.

"I am honored to receive this award from the Best of Los Angeles Award," says Lipps. "Everyone here at Lipps LA works hard every day to deliver the best results for our clients."
