A ranch-style design, low-maintenance living and convenient Fox Valley location created the perfect combination for Pam and Paul Wieland, who purchased the Oakland at Tanglewood Oaks in Aurora.

“The Oakland was just what we were looking for,” said Paul Wieland who moved to Tanglewood Oaks with his wife in June. The couple downsized from a traditional two-story in Batavia after realizing that with their children grown and out of the house, they only used “about half the rooms.” They looked at resales, which needed to too many upgrades, and custom homes before finding the best fit with Tanglewood Oaks.

With 2,134 square feet, the Oakland is actually about the same square footage as the Wieland’s old house, but the space is centered in the areas they live in most—like the Great Room that opens to the kitchen and adjoining breakfast nook. The Oakland also has two bedrooms plus an office, which the Wielands use when occasionally working at home, along with two full baths, a formal dining room, basement, and two-car garage.

In addition to the open floor plan, Pam appreciates the spacious gourmet kitchen. “It’s what I always wanted,” she said.

Other favorite features in the home are the fireplace and hardwood floors throughout, which are not only beautiful but practical for the couple’s two dogs. They added top-of-the-line wood banisters to the staircase leading to the basement for an upscale touch, as well.

The Oakland ranch home design was not the only thing that attracted the Wielands to Tanglewood Oaks. They also were drawn to the neighborhood and its wooded surroundings.

“We first stopped out here in October and the property was gorgeous,” said Paul Wieland. The couple was one of the first to visit the community once K. Hovnanian® Homes® opened for sale. “The leaves were changing in the neighboring forest preserve and the landscaping was well manicured and lush. We knew we had to come here.”

In fact, they liked the wooded environment so much they chose a homesite that backs up to the forest preserve.

Low maintenance was another deciding factor for the Wielands when choosing Tanglewood Oaks as their new home.

“A few years ago, we hired a lawn care service and got accustomed to not having to worry about cutting the grass and keeping up the landscape,” said Wieland. “At Tanglewood Oaks, we live among beautiful grounds and have someone who takes care of them. What more could we ask for?”

The Wielands moved to Tanglewood Oaks from Batavia, only about eight miles away. Both Pam and Paul had just turned 60 and wanted to settle into a place and stay for a while as they approached retirement. “A block away is the Gilman Trail, a popular bike path. And other amenities are nearby, too,” said Wieland. “But because of the forest preserves, it feels like we’re living in the country.”

Since moving in June, The Wielands have gotten to know many of their new neighbors.

“This is a close-knit community and the group of people who already lived here were very welcoming,” he said.

NATURE: Study Creates Cell Immunity to Parasite That Infects 50 Million

Collaboration between University of Colorado Cancer Center and University of Virginia looks past antibiotics and sanitation to a third strategy in the control of infectious disease: Adjusting the landscape of the human body to remove the mechanism that allows pathogens to kill.

There are two common approaches to protecting humans from infectious disease: Targeting pathogens and parasites with medicines like antibiotics, or dealing with the conditions that allow transmission. A paper published today in the journal Nature Scientific Reports demonstrates the effectiveness of a third strategy: Adjusting the landscape of the human body to remove the mechanism that allows pathogens to cause disease. The discovery is the result of serendipity and collaboration between high-level scientists in different fields.

"It was pure luck that I ended up on this paper," says Dan Theodorescu, MD, PhD, director of the University of Colorado Cancer Center. "Bill Petri and I had been social friends for years – Christmas parties, that kind of thing. When I was at Virginia it happened that we were on a recruitment committee together and the candidate was late, so we started talking."

His conversation with William A. Petri, Jr., MD, PhD, chief of the Division of Infectious Diseases & International Health at the University of Virginia led to the idea of applying an innovative cancer science technique to the study of infectious disease. With first author Chelsea Marie, PhD, postdoctoral researcher in the Petri Laboratory at Virginia, the group decided to silence genes in human cells to discover if the loss of any single gene would confer immunity to the parasite E. histolytica, which infects 50 million people and causes 40,000-110,000 deaths via severe diarrhea worldwide.

"Chelsea is a fearless experimenter. She took a library of cells that Dan had developed in his work with bladder cancer and then sequentially killed them with E. histolytica parasites," Petri says.

Specifically, the group used the technique called RNAi to create a library of bladder cancer cells with thousands of independent, silenced genes. Then they challenged these cultures with the parasite E. histolytica.

"We do this all the time in cancer research," Theodorescu says. "Commonly, we're looking for genes that, when silenced, will make cells more susceptible to chemotherapy."

In this case the analogue of chemotherapy was the infectious, dangerous pathogen.

"This amoeba is a cluster bomb – a voracious killer. In the back of my mind I was thinking the parasite was going to decimate the host cells no matter what we did with their genetics," Marie says.

For the vast majority of cells in this genome-wide screen, Chelsea Marie was correct; E. histolytica decimated many thousands of these independent cell cultures. However, a small number of cells seemed to resist the parasite. Was this the random chance of lucky survival or had silenced genes somehow offered immunity to these cells? To find out, Marie discarded the killed cells and retested the cells that had survived; again she infected these survivor cells with E. histolytica.

"It wasn't a fluke," says Marie. "We did this over nine generations of cells, each time selecting the cells that survived and then re-applying the parasite. Over these generations of selection, we saw the cultures becoming more and more enriched for cells lacking specific genes."

Using next generation sequencing, Marie identified the genes that conferred resistance and found that many were involved in managing the flow of potassium into and out of human cells. Specifically, the identified genes KCNA3, KCNB2, KCNIP4, KCNJ3, and SLC24A3 are involved in what is called potassium transport. A follow-up experiment showed that new intestinal cells treated with E. histolytica showed potassium efflux – the flow of potassium from inside a cell out through the cell wall – directly before cell death.

"We started to see a pretty clear line of reasoning," says Theodorescu. "The parasite was causing potassium efflux right before cell death and cells that happened to be unable to transport potassium didn't die."

To ensure that lack of potassium transport was, in fact, causing resistance to the parasite, the group reversed the direction of their experiments. Marie started with new cells and used drugs to block their ability to transport potassium. Blocking potassium efflux created cells that were resistant to E. histolytica.

"There is a clear need for new drugs targeting E. histolytica," Petri says. "Right now there is a single antibiotic that works against this parasite. We know that eventually the parasite will develop resistance to the antibiotic and at that point there's no plan B. This could be the plan B – targeting the human genes that enable the parasite to cause disease."

Marie is pushing forward. She recently learned from a mentor at John's Hopkins how to isolate stem cells from human tissue to grow what she calls "mini guts" to test therapeutics that may be useful in human patients. And technological advances make this study's general technique more efficient, allowing the use of what are called CRISPR libraries instead of RNAi screens.

"This is a major finding with translational implications for this infection that causes so many deaths worldwide, but also proof that this cancer-science approach can be used to explore genetic mechanisms of resistance in the field of infectious disease," Theodorescu says.

The field of infectious disease has been focused on the infection, targeting pathogens and their transmission. This study shows that in addition to characteristics of the parasite, mortality due to disease can be prevented by manipulating characteristics of the host. Reported by PRWeb 3 hours ago.

Three Pulmonary Hypertension Care Centers (PHCCs) have recently completed the Pulmonary Hypertension Association’s rigorous accreditation review process. The newly accredited centers join 26 U.S. healthcare facilities to receive the Pulmonary Hypertension Association’s prestigious accreditation since last year when the new review program launched.

Centers undergoing the accreditation process are healthcare facilities seeking to improve overall care and outcomes for people with pulmonary hypertension (PH). PH, specifically pulmonary arterial hypertension (PAH), is a debilitating disease of the lungs that affects the functioning of the heart and can lead to right heart failure.

In its nearly 25 years, PHA’s work has led to significant advances in PH care, including the availability of 12 FDA-approved treatments. However, the association continues to fight for earlier and more accurate diagnosis. With symptoms like shortness of breath and chest pains, PH patients often receive a misdiagnosis, going months believing they have asthma or another less-threatening disease. Many patients will see three or more physicians before they are properly diagnosed. Without treatment, the average survival is less than three years.

PHCC accreditation means a program demonstrates a dedication to making a proper diagnosis. It also means a program has the capacity to appropriately and comprehensively manage PH patients through a set of criteria established by the PHA’s Scientific Leadership Council and the PHCC Oversight Committee. The committee is comprised of members that are global leaders in the field of pulmonary hypertension.

Accreditation criteria reflect input from physicians, allied healthcare professionals, patients and PHA leadership. There are two types of accreditation: Centers of Comprehensive Care (CCC) and Regional Clinical Programs (RCP). Interested centers can apply online through the PH Care Centers Site Application Portal.

The three newly accredited PHCCs are:
1)    Allegheny General Hospital, Allegheny Health Network – Pittsburgh, Pa.
2)    Children’s Hospital Colorado Pulmonary Hypertension Program – Aurora, Colo.
3)    Comprehensive Pulmonary Hypertension Center (CPHC) – University Medical Center News Orleans, New Orleans, La.

As part of the PHCC accreditation process, PHA is developing the PHA Patient Registry (PHAR) of patients with PH. Accredited PH Care Centers and the patients they are managing will contribute data to PHAR, allowing researchers to track diagnostic and treatment patterns at a national level, ultimately demonstrating improvements in quality of care and quality of life of PH patients. Linking PHCCs through a national network will increase collaboration in the PH community, through clinical, quality improvement and investigative partnerships.

Confirmit Puts the Spotlight on Location-Based Customer Insight

Location and Beacon Triggering solution harnesses context-rich, in-the-moment feedback

Confirmit has developed a Location and Beacon Triggering solution which will enable users of Confirmit Horizons to harness mobile, location and beacon technology to better understand and act upon ‘in-the-moment’ customer behavior as part of a Voice of the Customer (VoC), Voice of the Employee (VoE) or Market Research (MR) program.

The increasing pressure faced by retailers and other customer-facing businesses to create dynamic shopping experiences that will encourage customers to return, either to browse and then buy online or to repeat purchase in-store, is a key factor behind the development of the solution.

“The popularity of mobile shopping apps and the willingness of shoppers to share their location, provided they get value in return, is expected to result in 72 percent of retailers implementing beacon technology within the next 5 years (Beaconstac). This means it is imperative that this technology becomes an integral part of our customer experience and MR solutions,” said Terry Lawlor, EVP of product management at Confirmit.

“Confirmit Horizons supports the creation of rules based on combinations of location events, allowing for selective triggering of ‘in-the-moment’ actions, such as requesting customer feedback. This will dramatically improve the ability of businesses to understand key customer behaviors, such as path to purchase or non-purchase and promotion conversion rates, for example.

“By offering an integrated location and beacon triggering solution as part of Confirmit Horizons, we enable our customers to correlate location-based data with other rich sources of customer data, such as purchase patterns and demographics, to deliver unparalleled customer and market intelligence,” Lawlor said.

The Confirmit solution captures location events through geo-fencing or beacon technology and identifies when the mobile device is near, has entered, or has exited a specific location, such as a store or branch, or is close to a key location such as an in-store display. By building rules based on combinations of these location events, customized and context-rich messages or surveys can be selectively sent to customers or panelists via a fully-branded app. The solution allows businesses to maintain complete control of when and how next-step actions are deployed.

As part of Confirmit Horizons, all data captured through the new solution is stored in Confirmit SmartHubTM, enabling location-based data to be combined with other sources of customer data to provide deep analysis of customer behavior.
